A simple, quantifiable, and accurate method for applying a noxious mechanical stimulus.

چکیده
I n animals, anesthetic requirements for a supramaximal noxious stimulus are usually determined using a hemostat applied to the tail or extremity (1,2). Although a generally accepted technique, the applied force is either not stated or unknown. Furthermore, when a noxious mechanical stimulus is applied to determine neurophysiological responses (e.g., dorsal horn activity), the force is often varied and must be accurately known. We sought to determine whether spring-activated A-clamps could be used to reproducibly apply a known force. The force (F) required to deform a spring is proportional to the spring constant (k). In the case of compressing or stretching a spring a certain distance (x), F = -kx. Springs in A-clamps apply force as the result of twisting along the longitudinal axis, with the force proportional to the amount of twisting. Additionally, adjusting the area of the A-clamp in contact with the tissue can vary the force applied per unit area of tissue.

FORMALIN AS A PERIPHERAL NOXIOUS STIMULUS CAUSES A BIPHASIC RESPONSE IN NUCLEUS PARAGIGANTOCELLULARIS NEURONS
The effects of formalin as a peripheral noxious stimulus on the activity of lateral paragigantocellularis nucleus (LPGi) neurons were examined. Spontaneous activity ofLPGi neurons was recorded after confirmation of their responsiveness to acute pain, and thereafter formalin (50 µL, 2.5%) was injected in the contralateral hindpaw. The response of the LPGi neurons was monitored for 60 min. A...
متن کاملEFFECTS OF FORMALIN AS A PERIPHERAL NOXIOUS STIMULUS ON THE NUCLEUS RETICULARIS PARAGIGANTOCELLULARIS NEURONS OF ANESTHETIZED RATS
In the present study, the effects of formalin as a peripheral noxious stimulus on spontaneously active units of the nucleus reticularis paragigantocellularis (PGi), a narrow region of the ventral pontine reticular formation, was examined in urethane anesthetized rats. Spontaneous discharge of the PGi neurons was variable, ranging from I to 37 spikes per second. Formalin as a chemical irrita...
متن کاملA comparison of pain measurement characteristics of mechanical visual analogue and simple numerical rating scales.
Numerical rating scales and mechanical visual analogue scales (M-VAS) were compared for their capacity to provide ratio scale measures of experimental pain. Separate estimates of experimental pain sensation intensity and pain unpleasantness were obtained by each method, as were estimates of clinical pain. Orofacial pain patients made numerical scale and VAS ratings in response to noxious therma...
متن کاملNoxious behaviors from subcutaneous injection of formalin and morphine tolerance effects on the responses of addicted male rats
Formalin as a chemical noxious stimulus evokes a biphasic pain that its second phase as a chronic inflammatory pain is similar to clinical pain. In this study, we used 40 NMRI male rats (250-300 g), and experiments were carried out considering ethical rules. Experiments were performed in normal and morphine addicted rats, the addiction was induced by Leung method. Behaviors of flinching and lic...
متن کاملThermoelastic Analysis of a Functionally Graded Simple Blade Using First-Order Shear Deformation Theory
In this article, the thermo-elastic behavior of a functionally graded simple blade subjected to the mechanical and thermal loadings is presented, applying a semi-analytical method and a variable thickness cantilever beam model. A specific temperature gradient is employed between the root and the edges of the beam. It is assumed that the mechanical and thermal properties are longitudinal directi...
